Description
Betulinic acid is a naturally occurring pentacyclic triterpenoid derived from the lupane skeleton, characterized by a 3β-hydroxy group, a carboxylic acid moiety at C-28, and a double bond between C-20 and C-29. It is commonly isolated from the bark of various plant species, including Syzygium claviflorum, Paeonia emodi, and Bowdichia virgilioides, where it serves as a prominent plant-derived bioactive.
Extensively studied in natural product pharmacology, betulinic acid exhibits a broad spectrum of pharmacological properties, including anti-HIV, antimalarial, anti-inflammatory, and antineoplastic activities. Mechanistically, it induces tumor cell apoptosis via mitochondrial membrane permeabilization, ROS-mediated signaling, and the inhibition of DNA topoisomerase (EC 5.99.1.3).
Due to its potent biological activity and relatively low toxicity, betulinic acid has been evaluated in clinical trials, particularly for conditions such as Dysplastic Nevus Syndrome, and continues to be investigated as a promising scaffold in the development of plant-derived therapeutic agents.
